General description
The Luciferase Assay System is an extremely sensitive and rapid reagent for quantitation of firefly luciferase in studying gene regulation and function. Linear results are observed under optimal conditions, even with high enzyme concentrations (over at least eight orders of magnitude of enzyme concentration), and low luciferase intensity (less than 10–20 moles of luciferase). Generally, 100-fold greater sensitivity can be achieved over the chloramphenicol acetyltransferase (CAT) assay. The Luciferase Assay Reagent generates light that is nearly constant for at least 1 minute and so the assay is compatible with measurement of luciferase using a single-tube luminometer or a multi-well plate luminometer with an auto-injector. Firefly luciferase catalyzes the oxidative carboxylation of luciferin, a reaction with the highest efficiency of any known bioluminescence reaction. The light emission from the reaction can be recorded using a luminometer. This kit provides a simple means for detecting luciferase activity in transfected eukaryotic cells. The assay ensures maximal sensitivity, consistent light output, as well as accuracy and consistency when handling multiple samples.
